于春秋两季种植大白菜与芜菁自交系杂交产生的F2群体,筛选与控制芜菁(Brassica campestris L.ssp.rapifera Matzg)肉质根直径基因紧密连锁的分子标记,检测控制肉质根直径的QTL数量、效应及其在图谱中的位置。试验结果表明:F2群体的肉质根直径大小偏向于芜菁,且呈连续性分布,表明该性状为数量性状,受多个基因控制。在春季F2群体中共检测到4个QTLs,分别位于A09、A07、A02和A03连锁群。在秋季群体中采用BSA法筛选到23个与控制肉质根直径基因连锁的标记,构建出A09连锁群片段,在其内检测到1个QTL位点(flesh root size 1),LOD值为8.19,贡献率为36.07%。该遗传位点在春秋两季群体中检测稳定,是一个新的主效QTL,可在今后分子标记辅助选择育种及基因克隆中加以利用。
The F2 population generated by crossing Chinese cabbage and turnip inbred lines in spring and autumn was screened for molecular markers closely linked to the control of diameter genes of fleshy roots of Brassica campestris L.ssp.rapifera Matzg. QTL quantity, effect and its position in the map. The results showed that the diameter of fleshy root of F2 population tended to be turnip, and the distribution was continuous, indicating that the trait was a quantitative trait controlled by multiple genes. Four QTLs were detected in the spring F2 population, which were located in A09, A07, A02 and A03 linkage groups respectively. In the autumn population, 23 markers linked to flesh root diameter genes were screened by BSA method. A09 linkage group fragment was detected, and a QTL locus (flesh root size 1) was detected, with a LOD value of 8.19. The contribution rate is 36.07%. The genetic locus is stable in spring and autumn seasons and is a new major QTL that can be used in future molecular marker-assisted breeding and gene cloning.
