目的研究白细胞介素-2、代谢型谷氨酸受体1亚型(mGluR1)和雌激素受体(ER)在中枢神经系统神经细胞的表达,证实它们共存的可能性。方法使用种属特异性1抗(山羊抗IL-2、兔抗mGluR1、小鼠抗ER血清)的免疫细胞化学三重标记技术,在相邻的两张连续切片(1张显示IL-2,另1张显示mGluR1和ER)上证实大鼠大脑皮质、延髓和脊髓内上述3种物质的共存。结果在上述部位的中枢神经元内可见IL-2、mGluR1和ER3种免疫反应性标记细胞。IL\|2免疫反应产物为棕褐色,位于胞浆内。相邻切片的mGluR1免疫反应产物为蓝黑色,位于胞膜;ER免疫反应产物亦为棕褐色,位于胞浆或核内。显示mGluR1和ER的同一切片上有mGluR1/ER双标细胞,双标细胞约占全部标记细胞的50%~60%。通过对相邻的两张切片的投影核对,证实存在mGluR1/ER/IL-2三标细胞,三标细胞占mGluR1/ER双标细胞的30%。结论在大鼠中枢神经系统内mGluR1、ER和IL-2可共存于同一个神经细胞,从而首次在细胞水平为神经免疫内分泌网络学说提供了形态学依据。
Objective To investigate the expression of interleukin-2, mGluR1 and ER in central nervous system neurons and to confirm the possibility of their coexistence. Methods Immunocytochemical triple labeling with species-specific 1 anti-goat anti-IL-2, rabbit anti-mGluR1 and mouse anti-ER serum was performed on two adjacent serial sections (1 for IL-2, 1 shows mGluR1 and ER) on the co-existence of these three substances in the rat cerebral cortex, medulla oblongata and spinal cord. Results IL-2, mGluR1 and ER3 immunoreactive marker cells were found in the central neurons of these sites. IL | 2 immune response products for the tan, located in the cytoplasm. Adjacent sections of the mGluR1 immune response product is blue-black, located in the membrane; ER immune response products are also tan, located in the cytoplasm or nucleus. The mGluR1 / ER double-labeled cells were found on the same section of mGluR1 and ER, and double-labeled cells accounted for about 50% to 60% of all labeled cells. Projections from two adjacent sections were confirmed by the presence of mGluR1 / ER / IL-2 triple-labeled cells, which accounted for 30% of the mGluR1 / ER double-labeled cells. Conclusion mGluR1, ER and IL-2 may co-exist in the same nerve cell in the central nervous system of rats, thus providing the morphological basis for the neuroimmunoendocrine network theory for the first time at the cellular level.
