目的:探讨脂源性炎症因子TNF-α,IL-6及IL-18在2型糖尿病大鼠视网膜的表达变化。方法:采用模拟人类膳食结构的长期高脂肪饮食24wk加小剂量STZ注射复制2型糖尿病大鼠模型8只,并以正常饮食组健康大鼠8只作为对照;应用免疫组织化学和原位杂交技术检测了TNF-α,IL-6及IL-18蛋白及mRNA在2型糖尿病大鼠视网膜的表达变化,并应用病理图像系统对结果进行定量分析。结果:TNF-α,IL-6及IL-18蛋白和mRNA在2型糖尿病组大鼠视网膜中均呈阳性表达;与正常组大鼠相比,其平均灰度、平均吸光度以及积分吸光度均有显著性差异(P<0.05)。结论:TNF-α,IL-6及IL-18在2型DM大鼠视网膜的表达增高。
OBJECTIVE: To investigate the changes of the retinal expression of lipogenic inflammatory cytokines TNF-α, IL-6 and IL-18 in type 2 diabetic rats. Methods: Eight long-term high-fat diet simulating human diet and STZ-injected STZ were used to make a model of type 2 diabetic rats. Eight healthy rats in the normal diet group were used as controls. Immunohistochemistry and in situ hybridization The expression of TNF-α, IL-6 and IL-18 protein and mRNA in the retina of type 2 diabetic rats were detected, and the results were quantitatively analyzed by pathological imaging system. Results: The protein and mRNA expression of TNF-α, IL-6 and IL-18 in retina of type 2 diabetic rats were all positive. Compared with normal rats, the average gray level, average absorbance and integral absorbance Significant difference (P <0.05). Conclusion: The expression of TNF-α, IL-6 and IL-18 in retina of type 2 diabetic rats increased.
