以水为提取溶剂对荞麦壳黄酮的提取工艺进行了优化,同时考察了黄酮得率与抗氧化活性的相关性。基于Plackett-Burman试验设计考察了各因素对荞麦壳黄酮得率及DPPH自由基清除活性的影响。试验结果表明:以黄酮得率为响应值的模型不显著,提取次数和料液比对DPPH自由基清除活性的影响显著(P<0.05)。最佳提取条件:料液比(g∶m L)1∶20,80℃热水提取时间20 min,重复提取3次,黄酮得率最高,为1.273%,且抗氧化活性较强。荞麦壳黄酮得率与DPPH自由基清除活性具有一定的相关性,相关系数为0.650 4。
The extraction of flavonoids from buckwheat hulls was optimized by using water as extraction solvent, and the correlation between flavonoid yield and antioxidant activity was also investigated. Based on the Plackett-Burman design, the effects of various factors on the yield of flavonoids from buckwheat hulls and DPPH radical scavenging activity were investigated. The experimental results showed that the model with the yield of flavone was not significant, and the effect of extraction times and the ratio of solid to liquid on DPPH radical scavenging activity was significant (P <0.05). The optimal extraction conditions were as follows: the ratio of solid to liquid (g: m L) 1:20, the extraction time of hot water at 80 ℃ for 20 min, extraction three times, the highest yield of flavonoids was 1.273% and the antioxidant activity was stronger. The yield of flavonoids from buckwheat hulls had some correlation with DPPH free radical scavenging activity, with a correlation coefficient of 0.650 4.
