[Objective]To find out the optimal extraction process for separating total flavonoids from Hypecoum leptocarpum Hook.f.et Thoms.and its application for determination.[Methods]Using total flavonoids as indicator,an orthogonal experiment was performed to optimize the extraction process by investigating the methanol concentration,material-liquid ratio and extraction time.[Results]The absorbance for total flavonoids was found peaked at 510 nm.The orthogonal experiment results indicated that the optimal extraction condition was A1B2C2,i.e.,methanol concentration of 50%,material-liquid ratio:1∶15(W/V,g/mL),ultrasonic extraction twice and 20 min for each extraction.Given these conditions,the total flavonoids contained in the Hypecoum leptocarpum Hook.f.et Thoms was about 48.63 mg/g.[Conclusions]The method is efficient,easy to operate,and yields accurate results.
