1目的制备并评价混合胶原型类风湿性关节炎(RA)大鼠模型。2方法将实验大鼠分为对照组、低剂量组和高剂量组,每组10只。分别对3组动物通过尾部注射生理盐水、0.1mg/mL混合胶原和0.4mg/mL混合胶原,1.0mL/只,间隔1周注射1次,共注射3次。末次注射1周后逐周观察并记录大鼠一般情况,并进行关节评分。末次注射3周后,处死大鼠,取右踝关节组织进行HE染色。3结果低剂量组和高剂量组关节腔炎细胞浸润清晰可见,且高剂量组炎细胞浸润较低剂量组严重。低剂量组和高剂量组关节评分在不同时期均高于对照组;低剂量组和高剂量组在末次注射2周和3周后关节评分均高于末次注射1周后。4结论对实验大鼠经过3次尾部注射混合胶原,成功建立RA动物模型,其组织学及临床表现与人类RA相似。
1 Objective To prepare and evaluate a rat model of mixed collagen-type rheumatoid arthritis (RA). 2 Methods The experimental rats were divided into control group, low dose group and high dose group, 10 rats in each group. Three groups of animals were injected with normal saline, 0.1 mg / mL mixed collagen, and 0.4 mg / mL mixed collagen, 1.0 mL per mouse, respectively, once a week for 3 times. One week after the last injection, the general condition of the rats was observed and recorded weekly, and the joints were scored. Three weeks after the last injection, rats were sacrificed and the right ankle joint tissues were stained with HE. 3 Results Low-dose group and high-dose group of arthritis inflammatory cell infiltration clearly visible, and high-dose inflammatory cell infiltration in the lower dose group was serious. The joint scores of low-dose group and high-dose group were higher than those of control group at different time points. The joint scores of low-dose group and high-dose group after 2 and 3 weeks after the last injection were higher than those after the last injection. 4 Conclusion Experimental rats after 3 times the tail mixed collagen injection, the successful establishment of animal models of RA, the histological and clinical manifestations and human RA similar.
