目的:观察蛇六谷提取物对体外生长的人肝癌HepG-2细胞和大鼠胶质瘤C6细胞增殖的影响,并探讨其可能的作用机理。方法:采用系统溶剂法初步提取分离蛇六谷不同有效部位,运用MTT法和生长曲线法观察不同浓度的各提取物对体外培养的HepG-2和C6细胞增殖的抑制作用,并以流式细胞仪检测其对两种细胞凋亡的影响。结果:蛇六谷醇提取物、石油醚萃取物和乙酸乙酯萃取物具有一定的抑制该两种肿瘤细胞增殖的作用,以石油醚萃取物的抑制作用最为明显,呈现较好的剂量-效应曲线和时间-效应曲线;流式细胞仪检测发现石油醚萃取物具有诱导HepG-2和C6细胞凋亡的作用。结论:蛇六谷石油醚萃取物和乙酸乙酯萃取物可能是该药抗肿瘤的有效部位。
OBJECTIVE: To observe the effect of the extract from snake six leaves on the proliferation of human hepatoma HepG-2 cells and rat C6 glioma cells in vitro and to explore its possible mechanism. Methods: Different effective fractions of snake six grains were initially extracted and separated by systemic solvent method. The inhibitory effects of different concentrations of extracts on proliferation of HepG-2 and C6 cells in vitro were observed by MTT assay and growth curve method. Flow cytometry Instrument to test its effect on both cell apoptosis. Results: The extracts of H. lucidum, petroleum ether extract and ethyl acetate extract could inhibit the proliferation of the two tumor cells to some extent. The petroleum ether extract showed the most obvious inhibitory effect and showed better dose-effect Curve and time-effect curve. The results of flow cytometry showed that petroleum ether extract could induce the apoptosis of HepG-2 and C6 cells. Conclusion: Shexianu petroleum ether extract and ethyl acetate extract may be the effective anti-tumor part of the drug.
