报告利用吖啶橙(Acridineorange)荧光染色技术对正常豚鼠和由顺铂致聋豚鼠的内耳毛细胞和螺旋神经节细胞的DNA及RNA进行观察。结果显示,正常内耳细胞核内DNA呈亮黄绿色荧光,胞质RNA呈桔红色荧光;核酸代谢障碍的胞核DHN呈浅黄色荧光,胞质RAN呈浅绿色或粉红色荧光。认为顺铂致螺旋神经节细胞内核酸代谢障碍是其致聋原理之一。同时表明,荧光染色技术对研究内耳细胞核酸含量变化,探讨药物耳毒性机理是一种简便、有效的手段。
The report uses acridine orange (Acridineorange) fluorescence staining of normal guinea pigs and cisplatin-induced deaf guinea pig hair cells and spiral ganglion cells DNA and RNA were observed. The results showed that the DNA in normal inner ear nucleus was bright yellow-green fluorescence and cytoplasmic RNA was orange-red fluorescence. The nucleus DHN of nucleic acid metabolism disorder was light yellow fluorescence and the cytoplasm RAN was light green or pink fluorescence. Cisplatin caused by spiral ganglion cell nucleic acid metabolism disorders is one of its principles of deafness. At the same time, it is indicated that fluorescence staining technique is a simple and effective method to study the change of nucleic acid content in inner ear cells and to explore the mechanism of ototoxicity.
