目的:探讨运动诱导心肌细胞HSP70表达的规律及其对心肌细胞的作用。方法:20只雄性SD大鼠分3组:对照组不训练;有氧训练组大鼠不负重,每天训练75min;过度训练组大鼠尾部负重(体重的5%),每天训练180min。每周训练5天,休息2天,共训练16周。取心肌组织进行HE染色观察和HSP70表达检测。结果:有氧训练组大鼠心肌纤维较对照组增粗,结构清晰,肌纤维和细胞核大小匀称,排列较对照组更为紧密整齐,染色均匀,未见异常改变。过度训练组大鼠心肌细胞的体积明显增大、混浊肿胀,肌纤维间隙增大,心肌纤维排列较对照组和有氧训练组松散紊乱,细胞间界限模糊,有部分肌纤维断裂,并在心肌细胞周围可见大量红细胞。过度训练组HSP70表达明显高于有氧训练组及对照组。结论:心肌形态结构在有氧训练后发生适应性改变,心肌细胞HSP70表达增加;过度训练后则发生病理性改变,心肌细胞HSP70表达较有氧训练组亦显著增加。
Objective: To investigate the regulation of HSP70 expression in cardiomyocytes induced by exercise and its effect on cardiomyocytes. Methods: Twenty male Sprague-Dawley rats were divided into three groups: the control group did not train; the aerobic training group did not bear the weight and trained 75 minutes per day; the over-training group rats tail weight (5% of body weight), trained 180 minutes daily. Training 5 days a week, rest for 2 days, training a total of 16 weeks. Take myocardial tissue for HE staining and HSP70 expression. Results: Compared with the control group, the myocardial fibers in the aerobic training group were thicker and the structure was clear. The size of muscle fiber and nucleus were symmetrical and arranged more closely than the control group. The dyeing was uniform and no abnormal changes were found. The volume of myocardial cells in overtraining group increased significantly, swelling of turbidity and the interspace of muscle fibers increased. The arrangement of myocardial fibers was more loose than that in control group and aerobic exercise group. The boundaries between cells were vague and some of the muscle fibers were broken. Can see a large number of red blood cells. Over-training group HSP70 expression was significantly higher than aerobic training group and control group. CONCLUSION: The morphological changes of myocardium are adaptive after aerobic exercise, and the expression of HSP70 in cardiomyocytes is increased. Pathological changes occur after over-training, and the expression of HSP70 in cardiomyocytes is significantly higher than that of aerobic exercise group.
