目的观察加味丹参饮预处理诱导大鼠缺氧/复氧心肌细胞热休克蛋白70(HSP70) mRNA的表达。方法将培养72小时的乳鼠心肌细胞随机分6组,空白组正常培养;血清对照组加50%大鼠血清培养;含药血清组加50%含加味丹参饮的药物血清培养;缺氧/复氧组予缺氧再给氧。缺氧预处理组、加味丹参饮预处理组先给予缺氧预处理和加味丹参饮预处理,24小时后再予缺氧再给氧。结果加味丹参饮预处理和缺氧预处理24小时后心肌细胞存活率显著高于缺氧/复氧组(P<0.01),乳酸脱氢酶(LDH)及肌酸激酶(CK)显著低于缺氧/复氧组(P<0.01),HSP70 mRNA表达显著高于缺氧/复氧组(P<0.01)。结论加味丹参饮预处理具有延迟保护作用,其机理与诱导细胞内HSP70 mRNA合成有关。
Objective To observe the expression of heat shock protein 70 (HSP70) mRNA in rats with hypoxia/reoxygenation induced by Jiawei Danshen decoction. Methods The neonatal rat cardiomyocytes cultured for 72 hours were randomly divided into 6 groups. The blank group was cultured normally; the serum control group was cultured with 50% rat serum; the drug-containing serum group was supplemented with 50% drug serum containing Jiawei Danshen decoction; hypoxia/ The reoxygenation group gave hypoxia and then oxygen. Hypoxia preconditioning group and modified Danshen drinking pretreatment group were given hypoxia preconditioning and modified Salvia miltiorrhiza pretreatment, and then hypoxia and then oxygenation 24 hours later. Results The survival rate of cardiomyocytes after pretreatment with Jiawei Danshen drinking and hypoxia preconditioning was significantly higher than that of the hypoxia/reoxygenation group (P<0.01), and the lactate dehydrogenase (LDH) and creatine kinase (CK) were significantly lower In the hypoxia/reoxygenation group (P<0.01), HSP70 mRNA expression was significantly higher than that in the hypoxia/reoxygenation group (P<0.01). Conclusion Modified Salvia miltiorrhiza preconditioning has a delayed protective effect and its mechanism is related to induction of intracellular HSP70 mRNA synthesis.
