目的评价1,3-丁二烯(BD)作业工人外周血淋巴细胞遗传物质的损伤。方法收集个人职业史、年龄、性别、吸烟和饮酒状况等信息,气相色谱法检测作业环境的BD浓度,应用外周血淋巴细胞胞质分裂阻滞微核(CBMN)检测法和彗星试验,评价180名BD作业工人和58名非职业暴露对照人群外周血淋巴细胞染色体和DNA损伤水平。结果作业区空气中BD浓度为1.80(0.59~2.76)mg/m3。BD作业工人外周血淋巴细胞微核率(CBMN)、核质桥率(NPB)、核芽率(NBUD)和Olive TM分别为(6.76±4.99)‰、1.00(0.00~4.00)、2.00(0.00~7.00)和4.64(3.50~5.98),均高于非暴露组(3.10±2.65)‰、0.00(0.00~2.80)、1.00(0.00~5.00)和2.34(0.82~3.93),差异有统计学意义(P<0.01)。按BD作业工龄将180名BD作业工人分为3组:1~年、14~年和20~年组,校正了年龄、性别、吸烟和饮酒后,BD作业工人CBMN率有随作业工龄的增加而增加的趋势。结论在现有BD暴露水平下,外周血淋巴细胞胞质分裂阻滞微核法和彗星试验均能检出BD作业工人的遗传物质损伤,并有随作业工龄的延长而增加的趋势。
Objective To evaluate the damage of peripheral blood lymphocyte genetic material in 1,3-butadiene (BD) workers. Methods Personal information such as occupational history, age, gender, smoking and drinking status were collected. The BD concentration in the working environment was detected by gas chromatography. CBMN assay and comet assay were used to evaluate the BD Chromosome and DNA damage levels in peripheral blood lymphocytes of BD workers and 58 non-occupationally exposed controls. Results BD concentration in the air of work area was 1.80 (0.59 ~ 2.76) mg / m3. The CBMN, NPB, NBUD and Olive TM of BD workers were (6.76 ± 4.99) ‰, 1.00 (0.00-4.00), 2.00 (0.00 ~ (7) ~ 4.00) and 4.64 (3.50 ~ 5.98), respectively, which were significantly higher than those in non-exposed group (3.10 ± 2.65) ‰, 0.00 (P <0.01). According to the working age of BD, 180 BD workers were divided into 3 groups: 1 ~ year, 14 ~ year and 20 ~ year group. After adjusting for age, sex, smoking and drinking, the CBMN rate of BD workers increased with the working length of service Increasing trend. Conclusions Under the current level of BD exposure, the peripheral blood lymphocyte cytoplasmic cleavage block micronucleus test and comet assay can both detect genetic material damage of BD workers and increase with the extension of working length.
