【目的】探明猪圆环病毒2型(PCV2)干扰伪狂犬病疫苗(PRV)免疫效果的可能机制.【方法】用PCV2与PRV在体外分别单接种和共接种仔猪肺泡巨噬细胞(AM),在接种后不同时间用荧光定量PCR技术检测PRV载量与共刺激分子CD80GCD86m RNA水平的动态变化.【结果】PRV组上清中PRV载量在接种后都显著(P<0??05)高于PCV2+PRV组.与对照组相比,在接种后48h内,PRV组CD80m RNA水平在接种后增加并在48h时达到显著水平,而PCV2组与PCV2+PRV组的CD80 m RNA水平都显著减少;在接种后48h内,CD86m RNA水平在PRV组都显著增加,在PCV2+PRV组则都显著减少,但在PCV2组仅在接种后48h显著减少.【结论】PCV2能够减少猪AM中PRV载量与CD80GCD86基因转录,这可能是PCV2抑制PRV免疫效果的机制之一.
【Objective】 To explore the possible mechanism of PCV2 interfering with the immune effect of PRV vaccine.Methods PCV2 and PRV were used to inoculate and co-inoculate piglets with alveolar macrophages (AM) , And the dynamic changes of PRV load and costimulatory molecule CD80GCD86m RNA level were detected by fluorescence quantitative PCR at different time after inoculation.Results The PRV load in supernatant of PRV group was significantly higher after vaccination (P <0 05) In PCV2 + PRV group.Compared with the control group, CD80m RNA level in PRV group increased at 48h after inoculation and reached a significant level at 48h, but CD80mRNA level in PCV2 group and PCV2 + PRV group was significantly higher than that of control group , But decreased significantly in PCV2 + PRV group, but decreased significantly in PCV2 group 48h after inoculation.Conclusion PCV2 can reduce the expression of PRV in pig AM And CD80GCD86 gene transcription, which may be one of the mechanisms of PCV2 inhibition of PRV immune effect.
