通过根癌农杆菌EHA105介导转化草莓叶片,在卡那霉素选择压下连续分化选择、扩繁和生根培养,获得了26株转乙烯受体FaEtr2反义基因植株。PCR和Southern blot检测证明,该基因已经整合草莓染色体中。Northern分析表明,转入的反义基因对靶基因都有部分的抑制。对果实乙烯释放量测定表明,转基因草莓乙烯释放量降低。
Through Agrobacterium tumefaciens EHA105 transformation of strawberry leaves, continuous selection, propagation and rooting were carried out under kanamycin selective pressure, and 26 FaEtr2 antisense transgenic plants were obtained. PCR and Southern blot showed that the gene has been integrated in strawberry chromosome. Northern analysis showed that the introduced antisense gene partially inhibited the target gene. The results showed that ethylene release from transgenic strawberry decreased.
