目的:采用超高效液相色谱-高分辨飞行时间质谱(UPLC-TOF-MS)对乐脉颗粒中的化学成分进行鉴定。方法:采用Phenomenon Luna C18色谱柱(250 mm×4.6 mm,5μm);流动相为甲醇-水,梯度洗脱;流速1.0 m L/min;ESI-MS检测采用正离子模式。通过软件计算未知物峰的精确分子量,与处方中7味药材的化学成分进行比对,实现了对乐脉颗粒中未知化合物的鉴定。结果:共鉴定出化合物23个,分别为:枸橼酸、没食子酸、丹参素、阿魏酸、绿原酸、羟基红花黄色素A、丹参酮ⅡA、芍药苷、异阿魏酸、紫草酸、槲皮素、红花黄色素A、迷迭香酸、原儿茶醛、丹酚酸B、丹酚酸A、藁本内酯、芦丁、木香烃内酯、α-香附酮、去氢木香内酯、D-儿茶素、丹皮酚。结论:UPLC的高分离度结合飞行时间质谱仪的超高分辨率为中药复方化学成分的研究提供了一种快速有效的方法,为乐脉颗粒物质基础研究奠定了基础。
OBJECTIVE: To identify the chemical constituents of Lemai granules by ultra performance liquid chromatography-high resolution liquid chromatography-time of flight mass spectrometry (UPLC-TOF-MS). Methods: Phenomenon Luna C18 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase was methanol-water with gradient elution. The flow rate was 1.0 m L / min. The positive ion mode was used for ESI-MS detection. The exact molecular weight of the unknown peak was calculated by software and compared with the chemical components of the seven herbs in the prescription to realize the identification of the unknown compounds in Lemai granules. Results: A total of 23 compounds were identified as citric acid, gallic acid, danshensu, ferulic acid, chlorogenic acid, hydroxysafflor yellow A, tanshinone Ⅱ A, paeoniflorin, isoferulic acid, , Quercetin, safflower A, rosmarinic acid, protocatechuic aldehyde, salvianolic acid B, salvianolic acid A, ligustilide, rutin, woodlocolactone, , Dehydrocostus, D-catechin, paeonol. CONCLUSION: The high resolution of UPLC combined with the ultra-high resolution of time-of-flight mass spectrometer provides a rapid and effective method for the study of the chemical constituents of traditional Chinese medicine compounds, which lays the foundation for the basic research of Lemai particulate matter.
