为研究压力对筛板细胞形态和糖蛋白合成的影响,对筛板细胞进行体外培养并施与一定压力,相差显微镜下观察筛板细胞形态学改变,并采用显微荧光光度计对筛板细胞合成层粘蛋白(LaminaLN)及纤维连接蛋白(fibrinectionFN)进行定量测定。结果在静压力(50mmHg)下,筛板细胞形态变扁,随加压时间延长,胞浆内空泡增多,线粒体肿胀、部分空泡化;胞浆内可见髓鞘样结构;加压3天的筛板细胞合成LN及FN分别为42.2±4.67、63.65±11.20荧光强度(AU),与对照组比较,差异呈显著性(P<0.05),加压5天的筛板细胞合成LN及FN分别为36.65±8.45、60.90±11.91AU,与对照组比较仍有不同程度增高。显示压力能促使筛板细胞形态异常和糖蛋白合成增加。
In order to study the effect of pressure on cell morphology and glycoprotein synthesis, the cells were cultured in vitro and subjected to a certain pressure.
