热休克蛋白27基因在食管鳞状上皮癌变过程中的表达

目的:研究食管鳞癌、癌旁及正常食管粘膜中热休克蛋白27(HSP27)基因的表达。方法:采用RTPCR方法,定量检测26例食管鳞癌、癌旁及正常食管粘膜中HSP27基因表达水平。结果:以HSP27/GAPDHmRNA灰度比值表示HSP27基因的相对表达量。食管鳞癌、癌旁(不典型增生)和正常食管粘膜中的HSP27/GAPDH灰度比值分别为0.81±0.27、0.90±0.24和1.22±0.39。HSP27基因表达水平在食管鳞癌和不典型增生食管粘膜中的表达量明显低于正常食管粘膜(P<0.05),而HSP27基因在食管鳞癌和不典型增生食管粘膜中的表达量差别无显著性(P>0.05)。食管鳞癌、不典型增生食管粘膜及正常食管粘膜中的HSP27基因表达在不同患者年龄、性别和淋巴结转移之间均无显著性差异(P>0.05)。结论:HSP27基因在食管鳞癌和不典型增生食管粘膜中表达与正常食管粘膜相比明显降低,提示HSP27基因在食管鳞癌的发生、发展过程中有不同程度的表达缺失。因此,提高HSP27基因在食管鳞癌中的表达将来可能会成为一种非常有效的生物学治疗手段。 Objective: To study the expression of heat shock protein27 (HSP27) gene in esophageal squamous cell carcinoma, paracancer and normal esophageal mucosa. Methods: RTPCR was used to detect the expression of HSP27 in 26 cases of esophageal squamous cell carcinoma, paracancer and normal esophageal mucosa. Results: The relative expression level of HSP27 gene was expressed by the ratio of gray level of HSP27 / GAPDHmRNA. The gray ratio of HSP27 / GAPDH in esophageal squamous cell carcinoma, paracancer (atypical hyperplasia) and normal esophageal mucosa were 0.81 ± 0.27, 0.90 ± 0.24 and 1.22 ± 0.39, respectively. The expression level of HSP27 in esophageal squamous cell carcinoma and atypical hyperplasia esophageal mucosa was significantly lower than that in normal esophageal mucosa (P <0.05), while the expression level of HSP27 gene in esophageal squamous cell carcinoma and dysplasia esophageal mucosa was not significantly different (P> 0.05). The expression of HSP27 in esophageal squamous cell carcinoma, atypical hyperplasia esophageal mucosa and normal esophageal mucosa had no significant difference (P> 0.05) between different age, gender and lymph node metastasis. Conclusion: The expression of HSP27 in esophageal squamous cell carcinoma and atypical hyperplasia esophageal mucosa compared with normal esophageal mucosa significantly reduced, suggesting that HSP27 gene in esophageal squamous cell carcinoma during the development of varying degrees of loss of expression. Therefore, improving HSP27 expression in esophageal squamous cell carcinoma in the future may become a very effective biological treatment.
