目的:观察番荔枝内酯去乙酰紫玉盘素(Desacetyluvaricin)对人肝癌细胞HepG2中细胞外信号调节激酶(extracellular signal regulated kinase,ERKs)表达水平的影响,探讨去乙酰紫玉盘素对ERK/MAPK信号传导通路的影响及其抗肝癌增殖的机制。方法:以12.52、5、50μg/ml的Desacetyluvaricin处理HepG2细胞,Western Blot检测ERK、p-ERK蛋白表达水平的变化,分析去乙酰紫玉盘素对ERK/MAPK信号通路的影响。结果:Desacetyluvaricin处理HepG2细胞后,ERK和p-ERK蛋白表达水平明显下降,并呈剂量依赖关系,其中Desacetyluvaricin以50μg/ml的浓度处理HepG2细胞时免疫印迹条带的发光强度最弱,ERK和p-ERK蛋白的表达水平最低。结论:去乙酰紫玉盘素抗肝癌增殖可能与其抑制ERK/MAPK通路有关。
Objective: To observe the effect of Desacetyluvaricin on the expression of extracellular signal regulated kinase (ERKs) in human hepatocellular carcinoma cell line HepG2 and to explore the effect of deacetylase on the expression of ERK / Effect of MAPK signal transduction pathway and its mechanism of antiproliferative activity on. Methods: HepG2 cells were treated with Desacetyluvaricin at 12.52, 5 and 50μg / ml. The expression of ERK and p-ERK protein was detected by Western Blot. The effect of deacetylase on ERK / MAPK signal pathway was analyzed. Results: After treatment with Desacetyluvaricin, the expression of ERK and p-ERK protein in HepG2 cells decreased significantly and in a dose-dependent manner. Desacetyluvaricin showed the weakest light intensity in HepG2 cells treated with 50μg / ml Desacetyluvaricin. The expression of ERK and p- -ERK protein expression level is the lowest. Conclusion: The anti-hepatocellular carcinoma cell proliferation may be related to its inhibition of ERK / MAPK pathway.
