目的对传统的单细胞凝胶电泳方法进行改良,建立了一种更加简单、可靠、易操作的方法。方法对常规载玻片和磨砂载玻片进行了筛选,对传统的制胶方法进行了改良,在实验的流程中增加水洗和脱水操作。结果常规载玻片的背景荧光强度较低。采用镀膜法制备底胶,灌胶法制备含细胞胶层可以很好地克服脱胶的现象。水洗和脱水步骤的加入增加了实验结果的可靠性。结论采用改良的实验方法节约了时间,且实验结果更加可靠。
Objective To improve the traditional method of single cell gel electrophoresis and to establish a more simple, reliable and easy-to-use method. Methods The conventional glass slides and frosted glass slides were screened, and the traditional gumming methods were improved. The washing and dewatering operations were added during the experiment. Results Conventional slides have a lower background fluorescence intensity. Prepared by the coating method at the end of plastic, plastic irrigation gel prep method can be very good to overcome the phenomenon of degumming. The addition of washing and dewatering steps increases the reliability of the experimental results. Conclusion The improved experimental method saves time and the experimental results are more reliable.
