目的 探讨TGF- β1 及细胞外基质在不同病变阶段主动型Heymann 肾炎肾小球内的表达情况。方法 建立AHN 模型, 用免疫组化SP法检测TGF- β1 及纤维连接蛋白、IV 型胶原在不同病变阶段Heymann 肾炎大鼠肾小球内的表达并用计算机图像分析系统进行定量分析。结果 自AHN 诱发后第4 周开始, 随TGF- β1 表达的上调, AHN 大鼠肾小球内细胞外基质成分纤维连接蛋白、IV 型胶原的含量也增加, 并随病变进展而明显增多。结论 TGF- β1 可能通过刺激AHN 大鼠肾小球上皮细胞产生过量细胞外基质而介导了AHN 病变的发生及发展
Objective To investigate the expression of TGF-β1 and extracellular matrix in glomeruli of active Heymann’s nephritis at different stages of disease. Methods The AHN model was established. The expression of TGF-β1, fibronectin and type IV collagen in the glomeruli of Heymann’s nephritis rats was detected by immunohistochemical SP method and quantified by computer image analysis system. RESULTS: From the 4th week after AHN induction, with the increase of TGF-β1 expression, the content of extracellular matrix fibronectin and type IV collagen in glomeruli of AHN rats increased, and increased with the progression of the lesion. Conclusion TGF-β1 may mediate the occurrence and development of AHN pathology by stimulating the extracellular matrix of glomerular epithelial cells in AHN rats
