目的 建立纯度较高的适于试验研究的人绒毛膜滋养层细胞 .方法 胰蛋白酶 - DNase联合消化法培养人绒毛膜滋养层细胞 ,间接免疫染色进行细胞鉴定 ,透射电镜观察细胞内部结构 .结果 倒置显微镜下 ,可见细胞呈不规则多角形 ,核大卵圆形 ,胞质透明 ,呈片状铺展生长 .角蛋白染色阳性率超过 95 % ,未检测到波型蛋白阳性细胞 ,表明不含污染细胞 .透射电镜观察示所获细胞有典型滋养层细胞结构 .结论 该法简便易行 ,可获得合乎试验要求的人绒毛膜滋养层细胞
Objective To establish human chorionic trophoblast cells suitable for experimental study.Methods Human chorionic trophoblast cells were cultured with trypsin - DNase and indirect immunostaining for cell identification, and the internal structure of cells was observed by transmission electron microscope Microscopically, the cells showed irregular polygons with large oval nucleus, transparent cytoplasm and flake spreading.The positive rate of keratin staining was over 95%, and no detectable wave form protein positive cells, indicating that the cells were free from contaminating cells Transmission electron microscopy showed that the cells were typical trophoblast cell structure.Conclusion This method is simple and easy to get in line with the requirements of human chorionic trophoblast cells
