目的探讨血管紧张素转换酶(Angiotensin Converting Enzyme,ACE)基因多态性与妊娠期高血压疾病发病的相关性。方法应用多聚酶链反应-限制性内切酶片段长度多态性(PCR-RFLP)技术,对90例妊娠期高血压疾病患者(妊高征组)、120例正常妊娠妇女(对照组)及90例妊娠期高血压疾病患者的母亲,其中41例患高血压(妊高征高血压母亲组)和49例无高血压(妊高征无高血压母亲组),进行ACE基因多态性的检测。结果妊高征组孕妇ACE基因的DD基因型及D等位基因频率均高于对照组,差异有统计学意义(P<0.05);妊高征高血压母亲组ACE基因的DD基因型及D等位基因频率均高于妊高征无高血压母亲组,差异有统计学意义(P<0.05);重度子痫前期孕妇与轻度子痫前期孕妇的ACE基因DD基因型分布频率比较有显著性差异(P<0.05);经相对危险度评估,ACE基因DD型和ID型的孕妇发生妊娠期高血压疾病的危险性增加,DD型个体患妊娠期高血压疾病的风险为正常孕妇的9.492倍。结论ACE基因I/D多态性可能与妊娠期高血压疾病发病有关,ACE基因中D等位基因可能是妊娠期高血压疾病的易感基因;ACE基因I/D多态性还可能影响到妊娠期高血压疾病病情的严重程度。
Objective To investigate the relationship between angiotensin converting enzyme (ACE) gene polymorphism and the incidence of hypertensive disorder complicating pregnancy. Methods Ninety pregnant women with gestational hypertension (PIH), 120 normal pregnant women (control group) and 90 healthy pregnant women (control group) were enrolled in this study. The polymerase chain reaction- restriction fragment length polymorphism (PCR-RFLP) Cases of pregnancy-induced hypertension in mothers, including 41 cases of hypertension (PIH mothers group) and 49 cases of non-hypertensive (PIH non-hypertensive mothers group), ACE gene polymorphism was detected . The risk of gestational hypertension in DD genotype was 9.492 Times Conclusion ACE gene I / D polymorphism may be related to the pathogenesis of hypertensive disorder complicating pregnancy. The D allele of ACE gene may be a susceptibility gene for hypertensive disorder complicating pregnancy. ACE gene I / D polymorphism may also affect Hypertensive disorders in pregnancy the severity of the disease.
