目的:研究复脉汤治疗充血性心力衰竭的作用及分子机制。方法:大鼠随机分成6组:假手术、模型、美托洛尔[5mg/(kg.d)]、复脉汤高、中、低剂量[26.7、17.8、8.9g/(kg.d)]。慢性心衰模型由结扎左冠状动脉诱导,药物从结扎当天开始灌胃给药,持续4周。结果:冠脉结扎后,动物体重增加百分率显著下降,心室重量指数显著升高,心肌酶谱AST,LDH,HBDH,CK及CKMB也显著升高,药物干预后体重下降得以缓解,同时升高的心室重量指数及心肌酶谱也显著下降。在模型组,RyR2及FKBP12.6的mRNA表达显著下降,药物干预后均显著上升。结论:复脉汤可通过改善肌浆网钙稳态治疗大鼠充血性心力衰竭。
Objective: To study the effect and molecular mechanism of Fumai decoction in treating congestive heart failure. METHODS: Rats were randomly divided into 6 groups: sham operation, model, metoprolol [5 mg/(kg.d)], Fumai Tang high, middle and low dose [26.7, 17.8, 8.9 g/(kg.d)] ]. The chronic heart failure model was induced by ligation of the left coronary artery and the drug was administered intragastrically from the day of ligation for 4 weeks. RESULTS: After coronary artery ligation, the percentage of weight gain in animals was significantly decreased, the ventricular mass index was significantly increased, and myocardial enzymes AST, LDH, HBDH, CK, and CKMB were also significantly elevated, and weight loss was relieved after drug intervention. Ventricular weight index and myocardial enzyme spectrum also decreased significantly. In the model group, the mRNA expression of RyR2 and FKBP12.6 decreased significantly, and they both increased significantly after drug intervention. Conclusion: Fumai Decoction can treat congestive heart failure in rats by improving sarcoplasmic reticulum calcium homeostasis.
