本文建立了用反相离子对 HPLC 法测定血浆中三种氟喹诺酮类广谱抗生素药物——诺氟沙星(Ⅰ)、阿米氟沙星(amifloxacin,Ⅱ)和依诺沙星(Ⅲ)的方法。样品制备取血浆样品1ml,加50μl 内标液(吡哌酸,50μg/ml),稍加涡旋后,再加50μl 高氯酸(60%,w/v),并同时涡旋30s,离心(10000×g)10min,取上清液20μl 注入色谱柱中进行测定。色谱条件不锈钢柱(100×2mm,id)匀浆填充5μm 的 ODS Hypersil 作为固定相;流动相为乙腈/水(25:75),其中含10mmol/L Na_2HPO_4、10mmol/L 十二烷基硫酸钠和5mmol/L 溴化四丁基铵,以磷酸调 pH 值至2.0;流速0.5ml/min;荧光检测280nm/418nm。
