目的:探讨枸杞总多糖(lycium barbarum polysaccharide,LBP)对2型糖尿病(NIDDM)大鼠细胞因子的调节作用。方法:采用高脂高糖饮食联合小剂量注射链尿佐菌素(streptozotocin,STZ)的方法制备NIDDM大鼠模型,随机选取40只均分为5组:模型组,LBP低(100 mg·kg-1)、中(200 mg·kg-1)、高(400 mg·kg-1)剂量干预组,格列苯脲干预组(阳性药物组);同时设立基础饲料喂养的正常对照组。Western blot法检测LBP对NIDDM大鼠脾脏组织中白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)表达的影响。结果:与正常对照组比较,NIDDM对照组大鼠脾脏组织中IL-1β,IL-6,TNF-α蛋白的表达均显著增高;以不同剂量的LBP干预治疗糖尿病大鼠4周后,各治疗组大鼠脾脏组织中IL-1β,IL-6,TNF-α的表达均显著降低。结论:LBP对NIDDM大鼠的免疫功能的调节作用可能与其抑制IL-1β,IL-6,TNF-α蛋白的表达有关。
Objective: To investigate the regulatory effect of lycium barbarum polysaccharide (LBP) on cytokines in type 2 diabetic rats. Methods: NIDDM rats were induced by high-fat and high-sugar diet combined with low dose streptozotocin (STZ) injection. 40 rats were randomly divided into 5 groups: model group, LBP low (100 mg · kg (200 mg · kg -1), high (400 mg · kg -1) dose intervention group and glibenclamide intervention group (positive drug group). Meanwhile, a normal control group was also established. The effect of LBP on the expression of interleukin-1β, interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in spleen of NIDDM rats was detected by Western blot. Results: Compared with the normal control group, the expression of IL-1β, IL-6 and TNF-α in the spleen tissue of NIDDM control group were significantly increased. After treatment with different doses of LBP for 4 weeks in diabetic rats, The expression of IL-1β, IL-6 and TNF-α in the spleen tissues of rats in the two groups were significantly decreased. CONCLUSION: The regulatory effect of LBP on the immune function of NIDDM rats may be related to its inhibitory effect on the expression of IL-1β, IL-6 and TNF-α.
