目的利用DNA条形码技术鉴定在入境航空器中截获的1只鼠类。方法提取截获鼠类的肝脏DNA,通过PCR方法扩增细胞色素C氧化酶亚基I(cytochromecoxidase I,CO I)基因并测序。将测序结果进行比对分析,从而鉴定物种。结果成功扩增到该鼠类样本的CO I基因,通过序列对比和系统进化分析,显示该鼠序列与黄胸鼠同源性为100%,鉴定为黄胸鼠。结论 DNA条形码技术在啮齿动物鉴定中应用良好,是一种简单、快速、高效的鼠类鉴定方法 。
Objective To identify a single rodent captured in an inbound aircraft using DNA barcode technology. Methods The murine liver DNA was intercepted and sequenced. The cytochrome c oxidase I (CO I) gene was amplified by PCR. The sequencing results were compared to identify species. Results CO I gene was successfully amplified into this murine sample. Through sequence comparison and phylogenetic analysis, the homology of this gene to Rattus flavipectus was 100%, which was identified as Rattus flavipectus. Conclusion DNA barcoding technique is well applied in rodent identification and is a simple, rapid and efficient method for the identification of rodents.
