目的研究丹参多酚酸盐对胶质瘤U251细胞增殖及凋亡的影响,并探讨其发生机制。方法应用CCK-8试剂盒检测丹参多酚酸盐对U251细胞增殖的影响,流式细胞仪观察丹参多酚酸盐对U251细胞周期及细胞凋亡的影响,q RT-PCR检测自噬基因Beclin-1的表达。结果丹参多酚酸盐可明显抑制胶质瘤U251细胞增殖,使细胞阻滞在G0/G1期,促进细胞凋亡,上调自噬基因Beclin-1 m RNA的表达。结论丹参多酚酸盐可抑制胶质瘤U251细胞生长,诱导细胞发生凋亡,促凋亡效应可能与自噬基因Beclin-1 m RNA表达增多有关。
Objective To study the effects of salvianolate on the proliferation and apoptosis of glioma U251 cells and to explore its mechanism. Methods CCK-8 kit was used to detect the effects of salvianolate on the proliferation of U251 cells. Flow cytometry was used to observe the effect of salvianolate on the cell cycle and apoptosis of U251 cells. The autophagy gene Beclin was detected by q RT-PCR -1 expression. Results Salviae miltiorrhizae polyphenols could significantly inhibit the proliferation of glioma U251 cells, arrest cells in G0 / G1 phase, promote apoptosis and up-regulate the expression of autophagy gene Beclin-1 m RNA. Conclusion salvianolate inhibits the growth of glioma U251 cells and induces cell apoptosis. The pro-apoptotic effect may be related to the increase of autophagy gene Beclin-1 mRNA expression.
