目的 :探讨进行膜受体免疫组化染色的标本制备时应采用的合适固定液浓度和固定时间 ,以提高反应的敏感性和稳定性。方法 :在不同固定液浓度和固定时间固定的脑切片 ,用免疫组织化学法显示人乙酰胆碱受体抗体( Ach Rab)与大鼠膜受体 Ach R的免疫结合反应。结果 :4 %多聚甲醛灌注固定 0 .5h且不经后固定组的染色结果最好 ,受体的免疫组化染色深 ,细胞数量多 ,突起长 ,其它组均不如这一组结果满意。结论 :用免疫组化检测膜受体时 ,应采用 4 %多聚甲醛灌注固定 0 .5h且不后固定的固定方法
OBJECTIVE: To investigate the appropriate concentration and time of immobilization for preparation of specimens for immunohistochemical staining of membrane receptors in order to improve the sensitivity and stability of the reaction. Methods: Immunohistochemical method was used to show the immunological binding of Ach Rab receptor to Ach R, a rat membrane receptor, in brain slices fixed at different concentrations of fixative solution and fixed time. Results: 4% paraformaldehyde perfusion fixed 0.5 h and without post-fixation group staining the best, immunohistochemical staining deep receptors, the number of cells, protuberances long, the other groups were not satisfied with the results of this group. Conclusion: Immunohistochemical detection of membrane receptors, should be used 4% paraformaldehyde perfusion fixed 0.5 h and not fixed after the fixation method
