目的:探讨P-gp与β-catenin和E-cadherin在结直肠癌发生发展过程中的作用及相互关系。方法:采用组织芯片、免疫组织化学技术检测135例结直肠腺癌及癌旁正常黏膜组织、89例腺瘤组织P-gp、β-catenin和E-cadherin蛋白表达情况,应用免疫荧光双重标记观察P-gp和β-catenin在腺癌组织定位情况。结果:P-gp在腺癌和癌旁正常黏膜的表达高于腺瘤(P<0.05),E-cadherin在癌旁正常黏膜和腺瘤的表达高于腺癌(P<0.05),在癌旁正常黏膜、腺瘤和腺癌组织中,β-catenin的细胞质及细胞核表达均呈逐级增高(P<0.05),P-gp与β-cate-nin细胞质/核表达与腺瘤不典型增生程度有关(P<0.05),E-cadherin表达与腺癌组织分化有关(P<0.05)。P-gp在腺瘤组织的表达与E-cadherin和β-catenin胞质表达呈正相关(P<0.05),在腺癌组织的表达与β-catenin胞质和胞核表达呈正相关(P<0.05),P-gp与β-catenin免疫荧光双重标记在腺癌组织呈共表达现象。结论:P-gp与β-catenin和E-cadherin之间存在密切联系,三者的检测对结直肠癌的早期诊断及耐药性评估有一定的帮助。
Objective: To investigate the role and relationship of P-gp, β-catenin and E-cadherin in the development of colorectal cancer. Methods: The expression of P-gp, β-catenin and E-cadherin protein in 135 cases of colorectal adenocarcinoma, adjacent normal mucosa and 89 cases of adenoma were detected by immunohistochemistry and tissue microarray. Immunofluorescence double labeling P-gp and β-catenin in adenocarcinoma tissue localization. Results: The expression of P-gp was significantly higher in adenocarcinoma and adjacent normal mucosa than in adenoma (P <0.05). The expression of E-cadherin in adjacent normal mucosa and adenoma was higher than that in adenocarcinoma (P <0.05) In normal mucosa, adenoma and adenocarcinoma, the expression of β-catenin in cytoplasm and nucleus increased progressively (P <0.05). The expression of P-gp and β-cate-nin in cytoplasm / (P <0.05). The expression of E-cadherin was correlated with the differentiation of adenocarcinoma (P <0.05). The expression of P-gp in adenoma was positively correlated with the cytoplasmic expression of E-cadherin and β-catenin (P <0.05), but positively correlated with the expression of cytoplasmic and nuclear β-catenin in adenocarcinoma ), P-gp and β-catenin double immunofluorescence staining in adenocarcinoma was co-expression phenomenon. Conclusion: There is a close relationship between P-gp and β-catenin and E-cadherin. The detection of these three drugs may be of help in the early diagnosis and drug resistance evaluation of colorectal cancer.
