目的应用支气管内细胞移植的方法建立大鼠肺肿瘤模型。方法40只雌性Wistar大鼠(平均体重169 g±10 g),用戊巴比妥钠腹腔注射麻醉大鼠,气管切开,超细导管支气管插管,接种肿瘤细胞(1×105~5×105)。病理学检查,计算肿瘤成活率。结果造模动物40只,存活39只。接种1×105~5×105个细胞各组动物的肿瘤成活率分别为60%、80%、90%和100%。结论应用支气管内细胞移植建立常位大鼠肺肿瘤模型的方法是可行的,其肿瘤成活率高,手术死亡率低,方法简单易行,易于重复。
Objective To establish a rat lung tumor model by intrabronchial transplantation. Methods Forty female Wistar rats (mean body weight 169 g ± 10 g) were anesthetized by intraperitoneal injection of pentobarbital sodium, tracheotomized and ultrafiltrated with bronchial cannula to inoculate tumor cells (1 × 105 ~ 5 × 105). Pathological examination, calculation of tumor survival rate. Results The model animals 40, 39 survived. Inoculation of 1 × 105 ~ 5 × 105 cells in each group of animals tumor survival rates were 60%, 80%, 90% and 100%. Conclusion The method of intrabronchial transplantation of human bronchial epithelial cells in establishing a model of rat lung tumor is feasible. The tumor survival rate is high and the mortality rate is low. The method is simple and easy to be repeated.
