目的:研究白细胞介素-2(IL-2)与单纯疱疹病毒1型(HSV-1)糖蛋白D(gD)基因共表达的核酸疫苗对小鼠免疫效果的调节作用。方法:构建表达HSV-1gD基因的核酸疫苗IRES-gD及共表达HSV-1gD与IL-2基因的重组质粒IRES-gD-IL-2,给BALB/c小鼠股四头肌注射,检测各组特异性抗体、中和抗体水平以及血清IgG2a/IgG1比值;并测定迟发型超敏反应(DTH)的强度;通过病毒攻击实验观察各组小鼠的生存情况。结果:IRES-gD-IL-2组产生的抗体及中和抗体滴度明显高于IRES-gD组。IRES-gD-IL-2免疫小鼠的IgG2a和IgG1含量及DTH反应均较IRES-gD组明显增高,但两组小鼠的生存率无显著性差异。结论:IL-2基因与HSV-1gD基因共表达可全面增强小鼠对核酸疫苗的免疫应答,尤其可增强细胞免疫效果。
AIM: To investigate the regulatory effect of nucleic acid vaccine co-expressing the gene of interleukin-2 (IL-2) and herpes simplex virus type 1 (HSV-1) glycoprotein D (gD) on immune responses in mice. Methods: The recombinant plasmid IRES-gD-IL-2 expressing HSV-1gD gene and the recombinant plasmid IRES-gD-IL-2 co-expressing HSV-1gD and IL-2 gene were constructed and injected into quadriceps femoris of BALB / c mice Group specific antibody, neutralizing antibody level and serum IgG2a / IgG1 ratio. The intensity of delayed-type hypersensitivity (DTH) was measured. The survival of mice in each group was observed by virus challenge. Results: The titer of antibody and neutralizing antibody in IRES-gD-IL-2 group was significantly higher than that in IRES-gD group. IgG2a and IgG1 levels and DTH responses in IRES-gD-IL-2 immunized mice were significantly higher than those in IRES-gD group, but there was no significant difference in survival rate between the two groups. CONCLUSION: The co-expression of IL-2 gene and HSV-1gD gene can enhance the immune response of mice to nucleic acid vaccine in general and can enhance the effect of cellular immunity in particular.
