刀豆蛋白A(简称Con A)可抑制SiO_2粉尘引起的巨噬细胞膜钾离子通透性的改变,并经电镜观察具有拮抗SiO_2致巨噬细胞损伤的作用。本实验通过观察Con A对SiO_2致红细胞溶血效应以及对红细胞膜内颗粒分布状态的影响,进一步探讨了细胞膜Con A结合部位在SiO_2致细胞膜损伤中的作用和意义。 材料与方法 雄性健康Wistar大鼠,断头取血,肝素抗凝,用pH 7.4的磷酸缓冲生理盐水冲洗离心,最终配制成1%(V/V)的红细胞是液备用。红细胞溶血测定:将不同量Sio_2加入红细胞悬液中,温育1小时后离心
ConA can inhibit the permeability of macrophage membrane permeability caused by SiO 2 dust, and can be observed by electron microscopy to antagonize SiO 2 -induced macrophage injury. In this experiment, the effects of Con A on hemolysis of erythrocytes induced by SiO 2 and the effect on the distribution of granules in erythrocyte membrane were observed, and the role and significance of Con A binding site in cell membrane damage induced by SiO 2 were further explored. Materials and Methods Male healthy Wistar rats were decapitated and anticoagulated with heparin. The samples were washed and centrifuged with phosphate buffered saline at pH 7.4. Finally erythrocytes were prepared as 1% (v/v). Red blood cell hemolysis assay: Different amounts of Sio_2 are added to the red cell suspension and incubated for 1 hour after centrifugation
