目的 探讨人重组生长激素正性肌力作用的机制。方法 利用Langendorff模型以及膜片钳技术研究心肌细胞内向钙电流。结果 受到人重组生长激素作用10min后,动作电位平台期延长,APD50、APD90分别由261.14±15.42ms至354.65±31.22ms和311.23±13.7至401.33±18.13ms。最大内向峰电流(Ica)由 2.03±0.09升至2.85±0.11PA,增加约40%。结论 人重组生长激素延长动作电位平台期,促进正常豚鼠心肌细胞L-Ca2+通道开放。
Objective To investigate the mechanism of positive inotropic effect of recombinant human growth hormone. Methods Langendorff model and patch-clamp technique were used to study the inward calcium current in cardiomyocytes. Results After treated with recombinant human growth hormone for 10 minutes, the plateau of action potential prolonged. The APD50 and APD90 were from 261.14 ± 15.42ms to 354.65 ± 31.22ms and 311.23 ± 13.7 to 401.33 ± 18.13ms, respectively. The maximum internal peak current (Ica) increased from 2.03 ± 0.09 to 2.85 ± 0.11PA, an increase of about 40%. Conclusion Human recombinant growth hormone prolongs the plateau of action potential and promotes the opening of L-Ca2 + channels in normal guinea pig cardiomyocytes.
