目的:研制出抗癌性缓释性栓塞制剂并初步观察临床疗效。方法:采用乙基纤维素包裹顺铂及硫酸钡制成微球。测定微球形态及缓释率。动物实验对顺铂微球的栓塞、缓释作用及毒性进行了观察。临床应用8例。结果:顺铂微球为规则球形,粒径为200±50μm,具有缓释性。动物实验表明微球注入后犬肝肾发生广泛坏死,微球可缓释出顺铂并且全身毒性小,临床应用疗效满意。结论:顺铂微球通过栓塞和缓释抗癌药物具有较强的抗癌作用,且副作用小,是较为理想的化学性栓塞制剂。
Objective: To develop an anti-cancer sustained-release embolization preparation and preliminary observation of clinical efficacy. Methods: The microspheres were made of ethyl cellulose encapsulated with cisplatin and barium sulfate. The microsphere morphology and sustained release rate were measured. In animal experiments, the embolization, sustained release and toxicity of cisplatin microspheres were observed. Clinical application of 8 cases. RESULTS: Cisplatin microspheres were regular spheres with a particle size of 200±50 μm and sustained release properties. Animal experiments showed that there was extensive necrosis of canine liver and kidney after infusion of microspheres. Microspheres could release cisplatin slowly and its systemic toxicity was small. Clinical efficacy was satisfactory. Conclusion: Cisplatin microspheres have good anti-cancer effects through embolization and sustained-release anti-cancer drugs, and have less side effects. It is an ideal chemical embolization agent.
