目的观察IL?4、TNF?α及IFN-γ诱导的MPM在抗弓形虫中的作用.方法采用3H-尿嘧啶(3H-U)特异性标记的弓形虫在小鼠腹腔巨细胞(Mouseperitonealmacrophage,MPM)内的增殖实验.结果与结论单独应用IL?4可部分抑制弓形虫在MPM内的增殖,且IL-4与IFN-γ在诱导MPM抗弓形虫的效应中具有相加作用,杭TNF-α中和抗体对IL?4诱导的弓形虫细胞内增殖抑制作用无影响;TNF?α与IFN-γ在诱导MPM抗弓形虫效应中存在协同作用,且其协同作用随TNF-α剂量的增高而明显增强.“,”Aim and Methods in vitro cultured MPM from C57BL/6 mice were treated with IL-4, TNF -α and IFN-β or a combination of these cytokines and ability to restrict growth of Toxoplasma gondii was then evaluted by H-U incorporation assay. Results and Conclusion IL- 4 alone inhibited the intracellular proliferation of Toxoplasma gondii only partially. No synergistic activity of IL-4 and IFN-β on the inhibition of Toxoplasma gondii prolifertion was found. The IL- 4 induced toxoplasmastatic activity was not affected by neutralizing antibodies against mouse TNF-α. Involvement of TNF-α in the induction of toxoplasmastatic activity was confirmed by our finding that TNF-α in combination with a nonactivating concentration of INF-β inhibited the intracellular proliferation of Toxoplasma gondii. TNF-α alone inhibited the intracellular proliferation of Toxoplasma gondii only slightly.
