对于细胞周期各不同时相辐射敏感性和突变发生的比较研究,一个多细胞有机体内的单细胞系统可以提供大量信息。单细胞阶段的受精卵——合子是最适宜的供试材料之一。对分析农艺性状的诱发突变,它也是适宜的。这是因为从诱变处理的卵细胞获得的个体,既没有二倍体选择,也没有嵌合体形成。作为研究工作的一个前提,阐明受精卵细胞第一次 DNA合成期以及其它细胞周期各时相的时间和持续期是必要的。本研究将确定水稻受精卵细胞与中央细胞第一次 DNA 合成期的时间和持续期,并确定受精卵的细胞周期各时相的持续期。~3H-TdR 掺入核的放射自显影观察表明:受精卵细胞第一次 DNA 合成期发生在第一次有丝分裂前约8小时,并表明受精作用完成后的合子 DNA 含量处于2 C 状态。
For a comparative study of radiosensitivity and mutation at different phases of the cell cycle, a single cell system in a multicellular organism can provide a wealth of information. Fertilized eggs at the single-cell stage - zygotes is one of the most suitable materials for testing. It is also suitable for the analysis of induced mutations in agronomic traits. This is because individuals obtained from the mutagenized egg cells have neither diploid selection nor chimerism formation. As a prerequisite for research, it is necessary to elucidate the timing and duration of the first DNA synthesis and other phases of the cell cycle of fertilized eggs. This study will determine the timing and duration of the first DNA synthesis of rice fertilized egg cells and central cells and determine the duration of each phase of the cell cycle of fertilized eggs. ~ 3H-TdR incorporation of nuclei by autoradiography showed that the first DNA synthesis of fertilized egg cells occurred about 8 hours prior to the first mitosis and showed that the zygotic DNA content in the fertilized state was in the 2C state.
