目的探讨FRMPD2基因表达下调对肺癌细胞生存、增殖以及黏附能力的影响。方法将FRMPD2sh RNA转染肺癌细胞SPC-A-1后,通过免疫荧光染色和Western blot检测FRMPD2的表达,并采用平板克隆形成实验、CCK-8增殖实验和细胞黏附实验测定转染细胞的生物学行为,分析FRMPD2表达相关的生物学效应。结果免疫荧光染色和Western blot结果均提示FRMPD2 sh RNA有效地下调了FRMPD2蛋白表达(<0.05)。体外实验显示,FRMPD2表达下调后细胞的集落形成率、细胞增殖能力和黏附率明显下降(<0.05)。结论 FRMPD2在肺癌细胞的生存、增殖和黏附等方面发挥重要作用。
Objective To investigate the effects of down-regulation of FRMPD2 gene expression on the survival, proliferation and adhesion of lung cancer cells. Methods FRMPD2sh RNA was transfected into SPC-A-1 lung cancer cells. The expression of FRMPD2 was detected by immunofluorescence staining and Western blot. The expression of FRMPD2 was detected by plate clone formation assay, CCK-8 proliferation assay and cell adhesion assay Behavior, analyze the biological effects associated with FRMPD2 expression. Results Immunofluorescence staining and Western blot showed that FRMPD2 sh RNA effectively down-regulated FRMPD2 protein expression (P <0.05). In vitro experiments showed that the cell colony formation rate, cell proliferation ability and adhesion rate were significantly decreased (P <0.05) after FRMPD2 expression was down-regulated. Conclusion FRMPD2 plays an important role in the survival, proliferation and adhesion of lung cancer cells.
