目的:对比研究兔多抗C-28和鼠单抗MET4在胰腺癌组织中检测MET蛋白表达的差异性和相关性。方法:采用免疫组化间接法研究C-28和MET4检测67例胰腺癌组织中MET表达情况,定量图像分析MET4和C-28的检测效能。结果:MET4和C-28检测胰腺癌的阳性率分别为70.1%和71.6%。两种抗体检测MET阳性率无显著差异(χ2=0.980,P=0.387)。相关分析显示两者无显著相关性(r=-0.127,P=0.307),在C-28未检出的病例中有84.2%(16/19)MET4检出,在MET4未检出的病例中有85.0%(17/20)C-28检出。在无MET蛋白表达的胰岛和正常胰腺上皮中,C-28有染色,而MET4无染色。结论:相对C-28抗体,MET4用于检测胰腺癌MET蛋白表达具有较高的特异性。
OBJECTIVE: To comparatively study the difference and correlation of MET protein expression in pancreatic carcinoma detected by polyclonal anti-C-28 and mouse monoclonal antibody MET4. Methods: The expression of MET in 67 pancreatic cancer tissues detected by C-28 and MET4 was studied by immunohistochemical indirect method. Quantitative images were used to analyze the detection efficiency of MET4 and C-28. Results: The positive rates of MET4 and C-28 in detecting pancreatic cancer were 70.1% and 71.6% respectively. There was no significant difference in the positive rate of MET between the two antibodies (χ2 = 0.980, P = 0.387). Correlation analysis showed no significant correlation between the two (r = -0.127, P = 0.307), 84.2% (16/19) of MET4 detected in C-28 undetected cases, and in MET4 undetected cases There were 85.0% (17/20) C-28 detected. In islet-free and normal pancreas without MET protein expression, C-28 was stained, while MET4 was non-stained. Conclusion: Compared with C-28 antibody, MET4 has high specificity for detecting MET protein expression in pancreatic cancer.
