目的探讨地塞米松早期干预对野百合碱(MCT)诱导肺动脉高压(PAH)大鼠肺血管活性物质的影响。方法成年雄性SD大鼠30只随机均分为对照(C)组、MCT模型(M)组、MCT+地塞米松(MD)组。建立MCT诱导PAH模型,应用放射免疫法观察血浆一氧化氮(NO)和内皮素1(ET-1)水平的变化。结果注射MCT 3周后,M组大鼠肺动脉平均压(mPAP)显著高于C组和MD组(P<0.05);与C组和MD组比较,M组血浆ET-1含量升高,NO含量降低(P<0.05);而MD组ET-l和NO含量与C组比较无统计学差异。结论地塞米松能抑制MCT诱导的PAH大鼠血浆中的ET-1含量升高和NO含量降低,从而改善内皮细胞功能,降低PAH。
Objective To investigate the effects of early dexamethasone on pulmonary vasoactive substances in rats with pulmonary arterial hypertension (PAH) induced by monocrotaline (MCT). Methods Thirty male SD rats were randomly divided into control group (C), MCT model group (M) and MCT + dexamethasone group (MD). The PAH model induced by MCT was established and the changes of plasma nitric oxide (NO) and endothelin-1 (ET-1) levels were observed by radioimmunoassay. Results After 3 weeks MCT, the mean pulmonary arterial pressure (mPAP) in M group was significantly higher than that in C group and MD group (P <0.05). Compared with C group and MD group, plasma ET-1 level was increased and NO (P <0.05). However, the levels of ET-1 and NO in MD group were not significantly different from those in C group. Conclusion Dexamethasone can inhibit the increase of ET-1 content and NO content in the plasma of PAH-induced PAH rats, thereby improving endothelial cell function and decreasing PAH.
