目的:研究磷脂酰肌醇3-激酶(PI3K)p110β蛋白在胃癌组织中的表达,并探讨其与预后的关系。方法:收集本院1996至2001年59例胃癌患者手术标本,应用PI3Kp110β多克隆抗体免疫组织化学方法检测其不同类型胃癌组织及癌旁正常胃组织中PI3Kp110β的表达。采用生存分析法分析其与预后的关系。结果:在59例胃癌组织中,PI3Kp110β阳性表达47例;在正常胃组织中其阳性表达13例(P<0.05)。肿瘤分化程度差的病例中PI3Kp110β阳性或强阳性表达明显高于肿瘤分化程度较好的病例(χ2=6.110,P=0.013);胃壁浸润较深(T3+T4)的病例中PI3Kp110β阳性或强阳性表达显著高于胃壁浸润浅(T1+T2)的病例(χ2=10.745,P=0.001);有淋巴结转移病例中PI3Kp110β阳性或强阳性表达高于无淋巴结转移的病例(χ2=4.511,P=0.034)。PI3Kp110β表达阳性或强阳性患者,其生存率显著低于PI3Kp110β表达弱阳性或阴性患者(P<0.01);且PI3Kp110β表达弱阳性患者其生存率也低于PI3Kp110β表达阴性患者(P<0.05)。结论:PI3Kp110β在低分化、浸润深及淋巴结转移的胃癌中高表达,PI3Kp110β的阳性表达对判断胃癌患者的预后有参考价值。
Objective: To study the expression of PI3K (PI3K) p110β protein in gastric carcinoma and its relationship with prognosis. Methods: Fifty-nine patients with gastric cancer from 1996 to 2001 were collected. The PI3Kp110β polyclonal antibody was used to detect the expression of PI3Kp110β in different types of gastric cancer tissues and adjacent normal gastric tissues by immunohistochemistry. Survival analysis was used to analyze the relationship with prognosis. Results: The positive expression of PI3Kp110β in 59 cases of gastric carcinoma tissues was 47 cases, while in normal gastric tissues, 13 cases were positive (P <0.05). The positive or strong positive expression of PI3Kp110β in cases with poorly differentiated tumors was significantly higher than those with well-differentiated tumors (χ2 = 6.110, P = 0.013). Positive or strongly positive PI3Kp110β in cases with deep gastric wall infiltration (T3 + T4) (Χ2 = 10.745, P = 0.001). The positive or strongly positive expression of PI3Kp110β in cases with lymph node metastasis was higher than that without lymph node metastasis (χ2 = 4.511, P = 0.034 ). The survival rate of PI3Kp110βpositive or strongly positive patients was significantly lower than that of PI3Kp110βwere weakly positive or negative (P <0.01). The survival rate of PI3Kp110βwere also lower than that of PI3Kp110β negative patients (P <0.05). Conclusion: PI3Kp110β is overexpressed in poorly differentiated, deep infiltrating and lymph node metastasis gastric cancer, and the positive expression of PI3Kp110β may be valuable in judging the prognosis of patients with gastric cancer.
