目的:研究赤芍总苷与黄芪多糖抑制血小板聚集的机制及其最佳配伍水平。方法:在对赤芍总苷与黄芪多糖分别采用不同剂量单味药实验的基础上,通过二次回归正交组合设计对赤芍总苷与黄芪多糖不同剂量配伍进行实验和分析。结果:赤芍总苷与黄芪多糖分别随着剂量的增加都能显著降低血小板的聚集率,赤芍总苷最佳剂量为125 mg.kg-1,黄芪多糖最佳剂量为300 mg.kg-1。赤芍总苷剂量和黄芪多糖剂量配伍与血小板聚集抑制率的关系模型为D=8.879+0.067 36 X1+0.164 75 X2+0.000 13 X1X2-0.000 542 X12-0.000 308 X22理论值与实际值能很好地吻合。结论:赤芍总苷与黄芪多糖对血小板聚集抑制有效,以88.908 mg.kg-1与284.3248 mg.kg-1水平配伍药效可能更好。
Objective: To study the mechanism of inhibition of platelet aggregation by total paeony glycoside and Astragalus polysaccharide and the best compatibility level. Methods: Based on the experiment of different dosages and single herbs of total glucosides of Radix Paeoniae Alba and astragalus polysaccharides, the orthogonal design of quadratic regression was used to test and analyze the different dosages of total glucosides of Radix Paeoniae Alba and astragalus polysaccharides. Results: TPG and Astragalus polysaccharide could significantly reduce the platelet aggregation rate with the increase of dose, the optimal dose of TPG was 125 mg.kg-1, the optimal dose of APS was 300 mg.kg- 1. Radix paeoniflorin total dose and Astragalus polysaccharide dose compatibility and platelet aggregation inhibition rate model for the D = 8.879 + 0.067 36 X1 + 0.164 75 X2 + 0.000 13 X1X2-0.000 542 X12-0.000 308 X22 theoretical and practical value can be very good To match. CONCLUSION: TPG and APS are effective in inhibiting platelet aggregation, and the pharmacodynamic efficacy may be better with 88.908 mg.kg-1 and 284.3248 mg.kg-1.
