目的:检测巨噬细胞游走抑制因子(MIF)在糖尿病大鼠心肌中的表达,探讨MIF在糖尿病心肌病变中的作用和用ISO-1[(S,R)-3-(4-hydroxypheny l)-4,5-dihydro-5-ISO-1xazole acetic acid methyl ester]干预后对MIF表达水平的影响及意义。方法:雄性SD大鼠30只,随机分为正常对照(NOR)组、糖尿病(DM)组和ISO-1组,每组各10只。用超声多普勒观察大鼠心脏左室舒张功能及免疫组织化学染色法检测各组大鼠心肌中MIF的表达。结果:与NOR组比较,DM组与ISO-1组舒张早期心室充盈速度最大值(E值)与舒张晚期心室充盈速度最大值(A值)的比值(E/A)明显降低(P<0.01),且DM组低于ISO-1组(P<0.01);NOR组心肌MIF表达水平显著低于DM组及ISO-1组(P<0.01),且ISO-1组低于DM组(P<0.01)。结论:MIF可能参与糖尿病心肌病的发生,其表达水平对预测心脏左室舒张功能障碍有重要价值,早期应用MIF抑制剂可能延缓糖尿病心肌病的发生,具有潜在治疗价值。
Objective: To detect the expression of macrophage migratory inhibitory factor (MIF) in the myocardium of diabetic rats, to explore the role of MIF in diabetic cardiomyopathy and to evaluate the effect of MIF on the myocardial infarction in diabetic cardiomyopathy with ISO-1 [(S, R) -3- ) -4,5-dihydro-5-ISO-lxazol acetic acid methyl ester] on MIF expression and its significance. Methods: Thirty male SD rats were randomly divided into normal control (NOR) group, diabetes mellitus group (DM) and ISO-1 group (n = 10). The left ventricular diastolic function was observed by Doppler ultrasound and the expression of MIF in myocardium was detected by immunohistochemical staining. Results: Compared with NOR group, the ratio of E / A to E / A in early diastolic ventricular filling velocity was significantly lower in DM group and ISO-1 group (P <0.01) ) In DM group were significantly lower than those in ISO-1 group (P <0.01). The level of myocardial MIF in NOR group was significantly lower than that in DM group and ISO-1 group (P <0.01) <0.01). CONCLUSION: MIF may be involved in the development of diabetic cardiomyopathy. The expression of MIF may play an important role in the prediction of left ventricular diastolic dysfunction in heart. Early application of MIF inhibitor may delay the development of diabetic cardiomyopathy, which may be of potential therapeutic value.
