目的探讨乙酰胆碱酯酶(acetylcholinesterase,AChE)在不同品系和不同性别淡色库蚊的变化,为应用生物化学方法检测蚊虫抗药性提供依据。方法以碘化硫代乙酰胆碱为底物,5,5′-二硫-双(2-硝基苯甲酸)为显色剂,测定单个蚊虫AChE,以残杀威为抑制剂测定AChE不敏感性。结果抗DDVP(Rd)和抗残杀威(Rp)品系淡色库蚊AChE活性水平高于敏感(S)品系,抗氯氰菊酯(Rc)品系淡色库蚊AChE活性与S品系相近。Rd、Rp品系淡色库蚊不敏感AChE个体频率高于S品系,Rc品系淡色库蚊不敏感AChE个体频率与S品系相近。S、Rd、Rp、Rc4个品系淡色库蚊3日龄雌雄成虫AChE活性均是雌性大于雄性。结论测定AChE可用来判断蚊虫抗药性状况,不同性别蚊虫应分别制定判断标准。作者简介:李士根(1966-),男,山东人,博士,副教授,主要从事医学寄生虫学教学与研究。E-mail:shigenli@yahoo.com.cn
Objective To investigate the changes of acetylcholinesterase (AChE) in Culex pipiens pallens (Culex pipiens) with different strains and genders, and to provide basis for the biochemical detection of Culex pipiens quinquefasciatus. Methods The AChE of single mosquito was determined with 5,1’-dithiobis-bis (2-nitrobenzoic acid) as substrate and thioacetyl iodide as substrate, and the insensitivity of AChE was determined by propoxur. Results The activity of AChE against Culex pipiens pallens in DDVP (Rd) and anti-prophylactic (Rp) lines was higher than that in susceptible (S) lines. AChE activity of Culex pipiens pallens was similar to that of S lines in resistant cypermethrin (Rc) lines. Rd, Rp strains of Culex pipiens pallens insensitive AChE individual frequency higher than S strains, Rc strains Culex pipiens pallens insensitive AChE individual frequency and S strains similar. S, Rd, Rp, Rc4 strains of Culex pipiens pallens 3-day-old male and female AChE activity were female than male. Conclusion The determination of AChE can be used to determine the status of mosquito resistance, and different gender should be developed respectively to determine the standard of mosquitoes.
