目的 探讨空间骨丢失的机理 ,研究模拟失重时人成骨样细胞细胞周期的变化。 方法 利用回转器模拟失重 ,观察测定了成骨样细胞增殖、周期的变化及其恢复作用和中药三花接骨散对细胞周期的影响。 结果 模拟失重作用后 ,成骨细胞增殖受到明显抑制 ,G1 期细胞显著增多 ,S与 G2 +M期细胞显著减少。模拟失重 12 h后再经 12 h以上正常生长 ,细胞周期可恢复至对照水平。三花接骨散能促进 G1 期细胞减少 ,S与 G2 +M期细胞增多 ,具有对抗失重效应 ,促进成骨细胞增殖的功效。 结论 模拟失重抑制成骨样细胞增殖 ,抑制细胞 G1 期向 S期的转化 ;模拟失重作用后细胞周期有一定的恢复作用 ;药物可以刺激细胞周期对抗模拟失重的效应。
Objective To investigate the mechanism of spatial bone loss and to study the changes of cell cycle of human osteoblast-like cells under simulated weightlessness. Methods The rotator was used to simulate the weightlessness. The proliferation and the cell cycle of osteoblast-like cells were observed and the recovery and the effect of Sanhua Decoction on cell cycle were observed. Results After simulated weightlessness, the proliferation of osteoblasts was significantly inhibited, the number of cells in G1 phase increased significantly, and the number of cells in S and G2 + M phase decreased significantly. Simulated weightlessness after 12 h and then by more than 12 h normal growth, the cell cycle can be restored to the control level. Sanhuajiegao can promote the reduction of G1 phase cells, S and G2 + M phase cells increased, with the effect of anti-loss of weight and promote the proliferation of osteoblasts. Conclusion Simulated weightlessness inhibits the proliferation of osteoblast-like cells and inhibits the transformation of cells from G1 phase to S phase. After simulated weightlessness, the cell cycle has a certain recovery effect. Drugs can stimulate the cell cycle to resist simulated weightlessness.
