利用融合蛋白表达载体pGEX-3X表达了戊型肝炎病毒第二(ORF2)和第三读码框(ORF3)区的两个抗原表位,并利用ORF2表位上游自然存在的限制性内切酶位点和ORF3下游通过PCR引入的限制性内切酶位点,使两个免疫表位区域形成嵌合。表达的三种抗原均溶于水。可利用商品化的谷胱甘肽-4B亲合层析柱得到纯化的抗原。经试用发现,基因重组抗原作为酶联免疫试剂的抗原用于检测血清中抗戊型肝炎病毒抗体,与新加坡进口试剂盒有高度的一致性。
The two antigenic epitopes of the second (ORF2) and the third reading (ORF3) regions of the hepatitis E virus were expressed using the fusion protein expression vector pGEX-3X and the restriction endonucleases naturally occurring upstream of the ORF2 epitope Site and a restriction enzyme site introduced by PCR downstream of ORF3 such that two immunological epitope regions form a chimera. All three antigens expressed are soluble in water. Purified antigens can be obtained using a commercial glutathione-4B affinity chromatography column.
