目的探讨辛伐他汀对人U251胶质瘤细胞增殖的影响。方法本实验分对照组和实验组,实验组的干预浓度分别0.2、1.0、2.0、4.0、8.0μmol/L5组,干预后培养24、48、72、96h,采用MTT比色法和流式细胞仪检测不同浓度及不同时间辛伐他汀干预后人胶质瘤细胞增殖活性和细胞周期的变化。结果辛伐他汀可明显降低人胶质瘤U251细胞活性,浓度到达1.0μmol/L时抑制作用明显。并和时间、浓度呈正相关,和对照组相比差异有统计学意义(P<0.05)。结论辛伐他汀对胶质瘤的增殖具有一定的抑制作用。
Objective To investigate the effect of simvastatin on the proliferation of human U251 glioma cells. Methods The experimental groups were divided into two groups: control group, experimental group and experimental group. The intervention groups were 0.2,1.0,2.0,4.0 and 8.0μmol / L5 respectively. After 24, 48, 72 and 96 hours of intervention, MTT assay and flow cytometry The proliferation and cell cycle of human glioma cells were detected by simvastatin with different concentrations and time. Results Simvastatin could significantly reduce the activity of human glioma U251 cells. The inhibitory effect was obvious when the concentration reached 1.0μmol / L. And the time and concentration were positively correlated with the control group, the difference was statistically significant (P <0.05). Conclusion Simvastatin has a certain inhibitory effect on the proliferation of gliomas.
