探讨氯化血红素用β-环糊精包合后的溶解度和体外溶出度 ,溶解度采用离心法 ,将氯化血红素及包合物用水配成饱和溶液 ,取适量以 40 0 0 r· m in- 1 离心 ,然后按分光光度法测定含量 ;溶出度采用桨法 (中国药典二部二法 ) ,以人工肠液为溶出介质 ,用分光光度法测定氯化血红素的含量。结果显示氯化血红素用β-环糊精包合后 ,溶解度和体外溶出度均有显著提高 ,制得的包合物水溶性好 ,体外溶出快 ,制备工艺简单易行。进一步证实包合物制备方法的可行性 ,同时也为氯化血红素加工成各种剂型开辟良好的前景。
To investigate the solubility and in vitro dissolution of hemin after inclusion with β-cyclodextrin, the solubility was determined by centrifugation, the hemin and the inclusion compound were mixed with water to form a saturated solution, take the appropriate amount of 40 0 0 r · m in 1 centrifuged, and then determined by spectrophotometry content; dissolution using paddle method (Chinese Pharmacopoeia two second method) to artificial intestinal fluid as the dissolution medium, spectrophotometric determination of hemin content. The results showed that the solubility and in vitro dissolution of hemin were significantly improved after inclusion with β-cyclodextrin. The obtained clathrate had good water-solubility and fast dissolution in vitro, and the preparation process was simple and easy. Further confirmed the feasibility of the inclusion complex preparation method, but also opened up a good prospect for the processing of hemin into various dosage forms.
