利用反相高效液相色谱建立同时测定新疆野山杏果肉中草酸、奎宁酸、酒石酸、苹果酸、抗坏血酸、乳酸、乙酸、柠檬酸、琥珀酸以及富马酸10种有机酸含量的方法。采用60%甲醇超声提取新疆野山杏果肉中有机酸,以Inertsil ODS-SP(4.6 mm×250 mm)C_(18)柱为分析柱,甲醇-(NH_4)H_2PO_4水溶液(p H=2.4)(3∶97,v∶v)为流动相,检测波长210 nm,流动相流速0.5 m L·min~(-1),柱箱温度30℃。结果表明,10种有机酸成分色谱峰分离度良好,方法回收率为93.20%~108.26%之间,RSD<3%;检出限为0.01~0.73μg/m L;定量限为0.03~2.39μg/m L。对野山杏果肉有机酸提取方法的考察表明超声提取比回流提取更方便快捷有效。
A method for simultaneous determination of 10 organic acids in oxalic acid, quinic acid, tartaric acid, malic acid, ascorbic acid, lactic acid, acetic acid, citric acid, succinic acid and fumaric acid in wild mountain apricot pulp was established by reversed-phase high performance liquid chromatography. The organic acids were extracted from the wild apricot pulp of the wild mountain by using 60% methanol. The chromatographic conditions were as follows: methanol - (NH 4) H 2 PO 4 aqueous solution (p H = 2.4) : 97, v:v) as the mobile phase, the detection wavelength was 210 nm, the mobile phase flow rate was 0.5 m L · min -1 and the oven temperature was 30 ℃. The results showed that the chromatographic peaks of the 10 organic acids were well resolved and the recoveries were between 93.20% and 108.26% with a RSD <3%. The limits of detection were 0.01-0.73μg / m L and the limits of quantification were between 0.03 and 2.39μg / m L. The study on the method of extracting organic acids from wild mountain apricot pulp showed that ultrasonic extraction is more convenient, quicker and more effective than reflux extraction.
