鱼白细胞介素-2是近年来新发现的一类细胞因子。根据B ird等发表的河豚IL-2基因序列设计一对特异性引物,通过RT-PCR方法扩增和克隆了红鳍东方鲀(Fugu rubripes)白介素-2的核苷酸序列。该cDNA序列由490nt组成,编码由149个氨基酸组成的前体蛋白。序列分析表明,红鳍东方鲀IL-2核苷酸序列和氨基酸序列与目前已知的其它物种IL-2的核苷酸及氨基酸的同源性分别位于34%~44%和24%~43%之间,N端存在长22个氨基酸的信号肽,含有形成2个链内二硫键的4个半胱氨酸。遗传进化分析表明,红鳍东方鲀IL-2在遗传进化上具有其独立性。
Fish IL-2 is a newly discovered class of cytokines in recent years. According to Bird et al., A pair of specific primers was designed for IL-2 gene of Puffer fish, and the nucleotide sequence of interleukin-2 of Fugu rubripes was amplified and cloned by RT-PCR. The cDNA sequence consists of 490 nt and encodes a precursor protein consisting of 149 amino acids. Sequence analysis showed that the nucleotide and amino acid identities of IL-2 nucleotide sequence and amino acid sequence of IL-2 were 34% -44% and 24% -43 %, A 22 amino acid signal peptide is present at the N-terminus and contains four cysteines forming two intra-chain disulfide bonds. Genetic analysis showed that the IL-2 gene had its own genetic evolution.
