目的建立同时测定了哥王中芹菜素和山奈酚含量的方法。方法采用HPLC法。色谱柱为Diamonsil C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-体积分数为0.4%磷酸-四氢呋喃(体积比40∶50∶10),流速为1.0 mL.min-1,检测波长为365 nm,柱温为35℃。结果芹菜素和山奈酚质量浓度分别在3.00~60.0 mg.L-1、0.347~6.94 mg.L-1内与峰面积呈良好的线性关系,平均回收率分别为100.2%(RSD=0.95%)和99.4%(RSD=1.0%)。结论该方法准确,重复性好,适用于了哥王药材的质量控制。
Objective To establish a method for the simultaneous determination of apigenin and kaempferol in Columbia king. Methods HPLC method. The column was a Diamonsil C18 column (250 mm × 4.6 mm, 5 μm) with a mobile phase of methanol-0.4% phosphoric acid-tetrahydrofuran (40:50:10 by volume) at a flow rate of 1.0 mL · min- 365 nm, column temperature 35 ℃. Results The concentrations of apigenin and kaempferol in the range of 3.00-60.0 mg.L-1.0.347-6.94 mg.L-1 showed a good linear relationship with the peak area. The average recoveries were 100.2% (RSD = 0.95%), And 99.4% (RSD = 1.0%). Conclusion The method is accurate and reproducible, which is suitable for the quality control of Columbia King.
