目的 :观察成年C5 7b1小鼠较长时间口服百草枯对其黑质纹状体系统的影响。方法 :将 48只成年C5 7b1小鼠随机分为 3组 ,每组 16只。MPTP组 :喂食MPTP 2 0mg·kg-1;百草枯组 :喂食百草枯 10mg·kg-1;对照组 :喂食等量生理盐水。每组小鼠每日喂食相应的毒物或生理盐水 ,共 2个月。观察小鼠的自发性行为、黑质部多巴胺能神经元以及纹状体中多巴胺、5 HT及其代谢产物含量的变化。结果 :MPTP 2 0mg·kg-1体重组及百草枯 10mg·kg-1体重组C5 7BL小鼠的自发性活动减少、纹状体中DA及其代谢产物含量降低、中脑黑质细胞数量减少。结论 :成年C5 7BL小鼠长期口服大剂量的百草枯可产生类似于帕金森病的行为 ,黑质纹状体也发生相应的生化病理改变
OBJECTIVE: To observe the effects of oral paraquat on nigrostriatal system in adult C5 7b1 mice over a long period of time. Methods: 48 adult C5 7b1 mice were randomly divided into 3 groups with 16 mice in each group. MPTP group: MPTP 20 mg · kg-1; paraquat group: paraquat 10 mg · kg-1; control group: the same amount of normal saline. Each group of mice fed the corresponding daily poison or saline, a total of 2 months. The changes of spontaneous behavior, dopaminergic neurons in substantia nigra and dopamine, 5 HT and their metabolites in striatum were observed. Results: MPTP 2 0mg · kg-1 body weight and paraquat 10mg · kg-1 body weight C5 7BL mice decreased spontaneous activity, decreased striatum DA and its metabolites, decreased number of midbrain substantia nigra cells . CONCLUSIONS: Long-term oral administration of high doses of paraquat to adult C5 7BL mice produces behavior similar to that of Parkinson’s disease, with corresponding biochemical and pathological changes also occurring in the nigrostriatal
